      ab226 staining CD90 / Thy1 in PC12 cells. The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab226 at 1/100 dilution (shown in green) and ab195889, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 594), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).

      Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

      Overlay histogram showing PC12 cells stained with ab226 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Triton X-100 for 15 min. The cells were then incubated in 1x PBS / 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (ab226, 1/500 dilution) for 30 min at 22°C.

      Isotype control antibody (black line) was Mouse IgG1 (monoclonal) FITC (ab18435) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.

      Acquisition of >5,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter.

    Thank you for contacting us and reporting the problems you have experienced in using ab226. The details provided will enable us to investigate this case and will provide us with vital information for monitoring product quality. Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab226. I would also appreciate if you can confirm some further details: 1. What washing steps were preformed? And with what buffers? 2. What volume of buffer was the cells suspended in for the primary antibody incubation? 3. Was the antibody kept in the dark at all times? 4. Was any analysis performed to detect the dead cells within the sample? I would suggest trying to use a higher concentration of primary antibody. The directions supplied with the datasheet are only guidelines and may need to be optimised for your particular experiment. I would suggest using a 1/10 dilution. It may also be worth trying a different blocking agent such as 3% BSA in PBS. Care should also be taken to keep the sample ice cold when preparing and I would suggest incubating the antibody with the cells (in the dark) for 1 hour at 4°C. Should the suggestions not improve the results, please do let me know. In the event that a product is not functioning in the species and applications cited on the product datasheet (and the problem has been reported within 6 months of purchase), we would be pleased to provide a free of charge replacement, credit note, or refund. We are happy to help customers find the most suitable products for their research purposes; however we are not specialized in this particular field and do not want to recommend a product that may be unsuitable. We encourage customers to consult the latest literature available through PubMed and other resources in order to find the most up-to-date information about their specific research interests. I found conflicting information about the expression of CD90 on NIH3T3 cells: No expression or expression of Thy1.2 (CD90.2). ab226 only recognizes Thy1.1 (CD90.1). I therefore suggest to use ab25672 (Anti-CD90 / Thy1 antibody [G7] (FITC)) which will bind CD90.1 and CD90.2. I could not find any reliable data on the expression of Ly6 on NIH3T3 cells either, but can confirm that ab25031 will bind to Ly6A.2 and Ly6E. I am sorry that I could not be more helpful, but I hope that the available literature in this area can provide some clarification.
