      Unfortunately we have not determined which specific HLA-A2 allele(s) this clone binds. However, it is clear from the paper you kindly provided that the clone binds HLA-A201. Unfortunately, we currently do not have any empirical data to indicate if this clone is specific just to the HLA-A201 allele or if detects other alleles.

      BATCH NUMBER 217168 ORDER NUMBER 168928 DESCRIPTION OF THE PROBLEM No signal. SAMPLE (Human) SupT1 cells (Human) Jurkat T cells PRIMARY ANTIBODY AbCam: Mouse monoclonal [BB7.2] to HLA A2(FITC). Used neat at 10uL per million cells Incubated for 1hr on ice or 1hr at Room Temp. Washed either once, twice or three times with 1xPBS DETECTION METHOD FACS POSITIVE AND NEGATIVE CONTROLS USED Negative Controls: Used unstained cells from both cell lines. Positive Controls: Used MHC-APC antibody* Initially used MHC-APC antibody*, but was hoping to use the AbCam to isolate specifically the HLA A2 population. ANTIBODY STORAGE CONDITIONS Ab received at room temp. Kept at 4C for ~2.5 weeks (while being tested) remainder was aliquoted and kept at -20C. SAMPLE PREPARATION Both cell lines are grown in 10% RPMI Ab/Amyc. Cells were resuspended in either solution buffers prior to Ab staining: 1xPBS or 1xPBS + 2%FBS NUMBER OF CELLS USED 0.5x10^6 (500,000) 1.0x10^6 (1,000,000) PERMEABILIZATION STEP N/A BLOCKING CONDITIONS Either not blocked (1xPBS) or blocked (1xPBS + 2%FBS) ~20min SECONDARY ANTIBODY N/A HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 4 HAVE YOU RUN A "NO PRIMARY" CONTROL?       Answer

      Thank you for filling out the questionnaire. Your protocol looks good. Do you incubate the cells with the antibody in the dark? FITC is sensitive to photo-bleaching. The only other recommendation I can make would be to try BSA instead of FBS for blocking, in case there are interactions with fetal serum proteins other than BSA that are blocking binding of the antibody to the HLA A2. The PBS-only control is a good idea but you may be losing the antibody to hydrophobic interactions with the tube you incubate in when you leave out blocking protein. The BSA is admittedly a long-shot solution. If you feel photo-bleaching is not a problem, then I can send you a free-of-charge replacement vial, or a refund or credit. Let me know what you would like.
