  • General notes

Reproducibility is key to advancing scientific discovery and accelerating scientists’ next breakthrough.

Abcam is leading the way with our range of recombinant antibodies, knockout-validated antibodies and knockout cell lines, all of which support improved reproducibility.

We are also planning to innovate the way in which we present recommended applications and species on our product datasheets, so that only applications & species that have been tested in our own labs, our suppliers or by selected trusted collaborators are covered by our Abpromise™ guarantee.

In preparation for this, we have started to update the applications & species that this product is Abpromise guaranteed for.

We are also updating the applications & species that this product has been “predicted to work with,” however this information is not covered by our Abpromise guarantee.

Applications & species from publications and Abreviews that have not been tested in our own labs or in those of our suppliers are not covered by the Abpromise guarantee.

  • Target

    • Relevance

Herpes simplex type 2 (HSV2) belongs to a family that includes HSV1, Epstein-Barr virus (EBV) and Varicella zoster (chicken pox) virus. HSV1 and HSV2 are extremely difficult to distinguish from each other. These viruses have a DNA genome, an icosahedral protein coat and are encased in a lipid membrane derived from the nuclear membrane of the last host. These viruses are capable of entering a latent phase where the host shows no visible sign of infection and levels of infectious agent become very low. During the latent phase the viral DNA is integrated into the genome of the host cell.

    Question

Does the antibody, Cat#AB20971 have cross reactivity with goat or rabbit antibodies. I'm using rabbit primary antibody and goat anti rabbit secondary as well as sheep anti HSV-2 on a mouse tissue, so, I'm wondering if they have cross reactivity with each other.

For further details, please see below:

I have used the sheep polyclonal FITC-HSV-2 Antibody (ab20971) and the result was very good. I'm quite satisfied with this antibody. The only matter is that I'm using this along with another antibody on a mouse tissue, that is rabbit anti mouse and then I add biotin goat anti rabbit to be able to amplify the signal by TSA kit. So, in recent immuno fluorescent studies I have seen such a cross reactivity between goat anti rabbit and sheep polyclonal anti HSV-2 antibody. I'm gonna make sure that whether this abcam sheep polyclonal anti HSV-2 antibody has cross reactivity with goat anti body.

    Answer

None of our primary antibodiesis checked against cross -reactivity with other IgGs.

However, we hardly come across complains in this direction.

Could you please send me the customer's protocol so I can have a look at it?

    Asked on May 10 2007

    Answer

The concentration of this product is 4 - 5mg/ml. The product is diluted to 5mg/ml and then vialed. Because other test methods can give different protein measurement results, the concentration is reported as 4 - 5mg/ml. The concentration of ab20971 is determined for the antibody after FITC-labeling. This is measured using OD280 with extinction coefficient E^0.1% = 1.4. The OD280 method measures total protein and therefore is measuring the antibody. However the FITC molecule is very small and does not significantly alter the concentration.
