    The V5 epitope tag is derived from a small epitope (Pk) present on the P and V proteins of the paramyxovirus of simian virus 5 (SV5). The V5 tag is usually used with all 14 amino acids (GKPIPNPLLGLDST), although it has also been used with a shorter 9 amino acid sequence (IPNPLLGLD).

    Thank you for the mail. As I mentioned in my previous mail the customer works with Drosophila BRAIN (fresh brain) that is fixed with 4% paraformaldehyde for 10 min. The customer does not perform any antigen retrieval. As this customer explained me this type of the staining that he performs is very similar to the staining of the cells.   Please advise if the following antibodies will be covered by Abcam's abpromise or you advise different choice. I am including other product under IHC-Fr application with PFA as fixative. Ab98795 is a secondary antibody; Abreview discount code is not valid for this product. The Abreview discount code for ab1274 and ab9116 can be provided however customer needs to explain how they have expressed tag conjugated protein in drosophila; please also forward the end user details.     Thank you for contacting us. These antibodies are against tags so species is not applicable for these products. "10 min in 4% paraformaldehyde and proceeds with the staining" is not enough information that we need for any recommendations. Please ask the customer to provide following info - Are they planning to use tissue sections or cells? What type of tissue section or cells they will be using e.g. Eye, Brain etc. - Are they planning Immunohistochemistry on frozen sections or paraformaldehyde sections thereby incubating sections with acetone or ethanol or treating sections with antigen retrieval method? - Now these antibodies are against tags so you may ask how they are expressing the tag in cells or tissues. Please provide the above information then only we can provide further confirmation.

    BATCH NUMBER 87891 ORDER NUMBER 26148 DESCRIPTION OF THE PROBLEM I have tried this antibody at 4 different concentrations ranging from 1:250 to 1:2000 and cannot see any staining by immunofluorescence. My staining protocol works for the Invitrogen monoclonal V5 and we have generated an antibody against the protein to which it is tagged and can see the distribution and increased expression by IF and western, respectively, but I see no staining with this polyclonal. If you have defined conditions for this antibody to work, please let me know. Thank you. SAMPLE primary epithelial cells PRIMARY ANTIBODY 1:250, 1:500, 1:1000, 1:2000 4oC overnight, three washes in TBS, blocked with BSA and 5%milk. POSITIVE AND NEGATIVE CONTROLS USED antibody against HA tagged protein, antibody against endogenous protein, Invitrogen monoclonal. SAMPLE PREPARATION 4% paraformaldehyde fixation and 0.5% triton permeabilization. Antibody added overnight at 4C. HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 4 HAVE YOU RUN A "NO PRIMARY" CONTROL?
