  • Additional notes

This peptide may be used for neutralization and control experiments with the polyclonal antibody that reacts with this product and FKBP 12, catalog ab2918. Using a solution with equal weights per unit volume of peptide and corresponding antibody will yield a solution with a large molar excess of peptide that is able to competitively bind the antibody.

  • Function
May play a role in modulation of ryanodine receptor isoform-1 (RYR-1), a component of the calcium release channel of skeletal muscle sarcoplasmic reticulum. There are four molecules of FKBP12 per skeletal muscle RYR. PPIases accelerate the folding of proteins. It catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides.
  • Sequence similarities
Belongs to the FKBP-type PPIase family. FKBP1 subfamily.
    Contains 1 PPIase FKBP-type domain.
  • Cellular localization
    Cytoplasm.
  • Information by UniProt

Customer reviews and Q&As

Answer

Thank you for your enquiry and for providing some details of your Western blot protocol. We would need further information of your assay and would be very grateful if you could answer the following questions. Your answers would help us identify the possible source of the problem. Please also provide the batch number and your order number and/or purchase order number. On what material are you testing the antibody in WB? • Species: • Cell extract or Nuclear extract: • Purified protein or Recombinant protein: The lysate • What lysis buffer was used: • What protease inhibitors were used: • What loading buffer was used: • Did you heat the samples: temperature and time: Electrophoresis/Gel conditions/ Transfer conditions • Reducing or non reducing gel: • Gel percentage : • Transfer conditions: Blocking conditions • Buffer: • Blocking agent: milk, BSA, serum, what percentage: • Incubation time: • Incubation temperature: Primary Antibody • At what dilution(s) have you tested this antibody: • What dilution buffer was used: • Incubation time: • Incubation temperature: • What washing steps were done: Secondary Antibody • Specification (in which species was it raised against)? • At what dilution(s) have you tested this antibody: • Incubation time • Wash steps: • Do you know whether the problems you are experiencing come from the secondary? Detection method ECl, ECl+, other detection method: Did you apply positive and negative controls along with the samples? Please specify. Optimization attempts • How many times have you tried the Western? • Do you obtain the same results every time e.g. are background bands always in the same place? • What steps have you altered? Could you please give us the Purchase Order Number and batch number corresponding to the product in question. The batch number will be printed on the vial label. If you are unable to give us the purchase order, please let us know the date you placed the order, or the name of the person who placed the order. Please also confirm that you ordered directly from Abcam rather than via one of our distributors. In this way we will be able to trace the date which the product was shipped and the batch number you received. We hope that we can help to solve this problem as quickly as possible.
