May be involved in the cytoskeletal organization of neuronal growth cones. Seems to be inactive as a PPIase (By similarity). Involved in the transport of early endosomes at the level of transition between microfilament-based and microtubule-based movement.
Belongs to the FKBP-type PPIase family. Contains 1 PPIase FKBP-type domain.
The PPIase FKBP-type domain seems to be inactive both for FK506-binding and enzymatic activity. The central coiled-coil region is responsible for association with early endosomes.

ab14432 staining FKBP135 in HeLa cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 3% BSA for 15 minutes at 20°C. Samples were incubated with primary antibody (1/200 in PBS) for 1 hour at 20°C. An Alexa Fluor®597-conjugated Donkey anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody. Cells labelled for FKBP15 displayed punctate staining, which was absent in cells treated with siRNA against FKBP15.

Antibodies: ab14432 used at 0.2 mcg/ml for WB in A and B and 2 mcg/mg extract for IP (B), ab14433 and ab14434 used at 2 mcg/mg extract for IP. Detection: Chemiluminescence with an exposure time of 25 minutes (A) and 10 minutes (B).
In figure B the Immunoprecipitation was done using ab14432 (Lane 3), ab14433 (Lane 4) and ab14434 (Lane 5) at 2µg/mg of extract.

ab14432 at 1/1000 staining mouse liver tissue sections by IHC-P. The tissue sections were paraformaldehyde fixed and blocked prior to incubation with the ab14432 for 24 hours. An HRP conjugated secondary antibody was used. The image shows staining in liver cells of a female mouse at 20X magnification.
