    Inquiry: Bonjour, J'ai utilisé cet anticorps pour la détection de FKBP51 sur lysat cellulaire (cellules humaines tumorales C4-2 et LnCap) et sur lysat de peau de souris. 30-50µg de protéine totale dilution anticorps primaire 1/1000 Anticorps secondaire GAR HRP Développé avec kit Dura J'obtiens deux bandes autour de 50kDa et deux autres (non spécifique?) à 15-20kDa Je voudrais savoir si les deux bandes à 50kDa correspondent à FKBP5 (isoformes?) et quel échantillon vous conseillez comme contrôle positif et négatif Merci d'avance

    Merci pour votre enquête.

    A ma connaissance, il n'existent pas des isoformes décrites de FKBP51. Il existent par contre des sites de phosphorylation. Un doublet de la bande autour de 50kD pourrait donc être du à phosphorylation différentielle. Je ne sais par contre pas à quoi les bandes entre 15 et 20kD pourraient correspondre.

    Pour exclure que les bandes observées sont des bandes non-spécifiques, je peux vous proposer d'investiguer les points suivants:

    1.) Les bandes pourraient être du bruit de fond du à l'anticorps secondaire reconnaissante des immunoglobulines endogènes ou du milieu de culture. Pour exclure cette possibilité, il suffit de faire à contrôle ou vous suivez le même protocole avec les mêmes échantillons etomettez l'anticorps primaire.

    2.) Les bandes observées entre 15 et 20kD pourraient être des produits de dégradation. Est-ce que vous avez ajouté des inhibiteurs de protéases au lysats? Je peux conseiller de préparer les lysats frais en ajoutant des inhibiteurs de protéases.

    Je serais ravi d'essayer de donner plus de conseils. Pour ceci je devrais regarder vos résultats en plus de détails. Si vous souhaitez ceci, veuillez me faire parvenirle plus d'informations possible sur le protocole que vous avez utilisé avec l'aide du questionnaire ci-joint.     Could you please let me know if your antibody anti-FKBP-51 cod.ab2901-10,could cross react with other immunophillins, in particular could the antibody recognize FKBP12? In our lysates, we detected FKBP51, but the antibody marked also a band under 16.5 MW .

    Answer

    Thank you for your enquiry. We did a BLAST search on the immunizing sequence and the only FKBP that came up was 51. We do not think this is 12 but it may be a degraded fragment from 51.
