    Target

    • Relevance

Fluorescein is a fluorophore commonly used to label proteins - protein-fluorescein conjugates are not usually susceptible to precipitation. In addition to its relatively high absorptivity, excellent fluorescence quantum yield and good water solubility, fluorescein has an excitation maximum of 494 nm that closely matches the 488 nm spectral line of the argon-ion laser, making it an important fluorophore for confocal laser-scanning microscopy and flow cytometry applications. Its fluorescence is pH sensitive and is significantly reduced below pH 7. Fluorescein emits most strongly between 500 and 550 nm, but it has a relatively broad emission spectrum reaching to over 600 nm. Several derivatives of fluorescein are commonly used, including FITC (fluorescein isothiocyanate), carboxylates and succinimidyl esters.

    Question

Today I have spoken on the phone to Danielle Miller about your Goat anti-fluoresceine AB6644. We do not have signal with the antibody, only an aspecific band. I have filled in a form via your website about the problems we have. Attached to this mail are two files with images of the blots we obtain: First:' blotabcam' which contains 3 samples and 2 markers. On the image should be clear that nr 1 is the negative control and 2 and 3 are the positive controls. This blot is detected with your antibody (1:1000)and as secondary antibody we have used donkey anti goat with peroxidase(1:5000) from Jackson Immuno Research. Second: ' blotbiogenisis.jpg' which contains 3 samples and 2 markers. On the image should be clear that nr 1 is the negative control and 2 and 3 are the positive controls. This blot is detected with rabbit anti fluoresceine conjugated with a peroxidase. The lot nr of our antibody is 8001. We would like to obtain another lot of the antibody to test again.
