1) The construct contains the full mouse FLVCR coding sequence
2) We have inserted a single copy of a MYC tag at the C-terminus of FLVCR
coding sequence. As the antibody is raised against an internal sequence of the
protein its unlikely that it could inhibit the binding of the antibody to
3) We didn’t use the brain as positive control but we have also tested the
liver from wild-type and FLVCR-null mice that we have generated in our
lab. Also in the liver, this antibody recognizes the same background band
at the expected/correct molecular weight, but does not show significant differences between wild type and knockout animals.
4) No, we haven’t run a “only secondary antibody” to see if any of the
non-specific bands are due to the secondary, because we have tested these secondary antibody with a lot of different cells and primary antibodies without obtaining a similar non-specific band
