  • Description
    Mouse monoclonal [5D7] to G-CSF
  • Host species
  • Tested applications
Suitable for: WB, Sandwich ELISA, Indirect ELISAmore details
  • Species reactivity
    Reacts with: Human
  • Immunogen

    Human recombinant granulocyte-colony stimulating factor.

  • Positive control
  • General notes

The neutralising bioassay was performed on myeloid leukemia G-NFS-60 cell line. G-CSF stimulates the proliferation of these cells in vitro. Briefly, human G-CSF was pre-incubated with monoclonal antibody 5D7 for 2 h at RT and then added to the cell suspension. Cells were cultured for 72 h. Cell proliferation was measured by MTT method. Polyclonal antibody to G-CSF and an irrelevant monoclonal antibody of the same isotype were used as controls.

This product was changed from ascites to tissue culture supernatant on 28/11/2017. Lot numbers higher than GR241046-2 will be from tissue culture supernatant. Please note that the dilutions may need to be adjusted accordingly.

Our Abpromise guarantee covers the use of ab9818 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB 1/1000 - 1/5000.
Sandwich ELISA Use a concentration of 1 µg/ml. Can be paired for Sandwich ELISA with Rabbit polyclonal to G-CSF (ab9691). For sandwich ELISA, use this antibody as Capture at 1µg/ml>ab9691 as Detection.
Indirect ELISA 1/2000 - 1/20000.

  • Function
Granulocyte/macrophage colony-stimulating factors are cytokines that act in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytes-macrophages. This CSF induces granulocytes.
  • Sequence similarities
    Belongs to the IL-6 superfamily.
  • Post-translational
O-glycan consists of Gal-GalNAc disaccharide which can be modified with up to two sialic acid residues (done in recombinantly expressed G-CSF from CHO cells).

Thank you for your patience in awaiting our reply. I have checked all of our monoclonal G-CSF antibodies and none of them have been epitope mapped, so we do not know where they bind. The polyclonal antibodies, of course, will bind many places. Please let me know if you have any further questions.

Thank you for your enquiry and interest in our products. As the datasheet indicates, this antibody (ab9818: anti-G-CSF antibody) is unconjugated so it is important to use a compatible secondary antibody which is AP-linked. Currently, we do not have AP-conjugated version of this antibody, however we have some biotin (ab17633 and ab74034) and HRP (ab106679) - labelled ones. I hope this helps and if I can assist further, please do not hesitate to contact me.

Thank you for your enquiry. I contacted the supplier of abs 9818, 9821, and 9822 and they were able to provide me with the following information. Ab9818 is non-neutralizing. It does not inhibit the biological activity of human G-CSF in vitro. The ability of ab9818 to inhibit proliferative activity of G-CSF was tested using G-NFS-60 cell lines. Neutralizing activity of abs 9821 and 9822 have not yet been tested. I am still waiting to hear back from the supplier regarding ab649. If you have any questions in the meantime, please contact us again.
