Target

  • Relevance

    Guanine nucleotide-binding proteins (G proteins) are involved as modulators or transducers in various transmembrane signaling systems. The Gs protein is involved in hormonal regulation of adenylate cyclase: it activates the cyclase in response to beta-adrenergic stimuli. Alternative splicing of downstream exons of the GNAS gene is observed, which results in different forms of the stimulatory G protein alpha subunit, a key element of the classical signal transduction pathway linking receptor-ligand interactions with the activation of adenylyl cyclase and a variety of cellular reponses. Multiple transcript variants have been found for this gene, but the full-length nature and/or biological validity of some variants have not been determined. Mutations in this gene result in pseudohypoparathyroidism type 1a, pseudohypoparathyroidism type 1b, Albright hereditary osteodystrophy, pseudopseudohypoparathyroidism, McCune-Albright syndrome, progressive osseus heteroplasia, polyostotic fibrous dysplasia of bone, and some pituitary tumors.

    Thank you very much for your interest in ab101736. I would recommend that when starting this procedure that you use an ice cold methanol or acetone fixation. Fixation times can vary from 1-10 minutes so it may be best to do a series of different times. Be sure to thouroughly rinse the samples in ice cold PBS following fixation. The advantage of using methanol or acetone is that they will permeablize the cells allowing you to skipthis step later. I would also recommend using a blocking solution containing 1% BSA in PBST or a 10% serum of the animal your secondary antibody was raised in. Block for at least 30 minutes at room temperatures. You will have to titer out the correct dilution of antibody to use for your experiments however a good starting point is 5ug/mL. Incubate at room temperature for 1 hour or overnight at 4C. I would like to direct you to our protocols and tips page for further ideas and detailed protcols. You may find these under the Immunostaining headingin the lower left hand column.     Hello, I am looking for an antibody that will recognize G protein alpha S subunit from mouse tissue (brain) for western blotting and immunochemistry. I need an antibody that will not recognize G alpha olf (or any other G protein alpha subunits). Are your Gs antibodies tested against other G protein subunits? The immunizing peptide only matches with mouse GNAS NP_034439.2 and GNASL NP_963910.1 (both GeneID 14683). I hope this information is helpful to you.
