Regulates osteoclast differentiation mediating the TNFRSF11A/RANK signaling. In allergic response, it plays a role in mast cells activation and degranulation through PI-3-kinase regulation. Also involved in the regulation of cell proliferation and hematopoiesis.

    Dear Sirs,

    I would like to query your technical support regarding the following antibodies:
    Ab62802,ab32365, ab78262 and ab92578.
    In order to detect changes in GAB2 and phospho-GAB2 levels we ordered several anti GAB2 and anti phospho-GAB2 antibodies from different sources.
    After trying several cell lysis conditions to minimize breakdown products, we settled for the cell lysis buffer by Cell signaling technologies (cat #3884 in combination with a protease inhibitor cocktail from Roche). Result where generated using the K562 cell line which is also mentioned in the datasheet of Ab62802,ab32365 and ab92578
    Using the competitors antibodies we are able to assure the availability of both GAB2 and phosphorylation site specific forms of phospho-GAB2.
    Unfortunately using the anti GAB2 antibodies ab62802,ab32365,ab92578 and ab78262 we are not able to get the desired results.
    Ab62802 does not yield a nice and clean band at the expected 80 to 90 KDa, instead we see a smear/aspecific binding to our membrane.
    The antibodies ab32365,ab92578 and ab78262 do not yield a band when using either lysates of K562 or IFN triggered Jurkat cells (according to the ab78262 datasheet).
    Since the competitors antibodies clearly show the availability of both GAB2 and pGAB2 in the K562 cell line we seriously doubt the effectiveness of the Ab62802,ab32365, ab78262 and ab92578 antibodies (please see attached pdf for a line up of the antibodies used (markings in blue are competitor products).
    Our question is, do you have any suggestion to alleviate our problems, we are somewhat at a loss here.

    In order to better understand the problem, I’d like to send you a questionnaire with some questions about the protocol used.

    Once we receive the completed questionnaire, we will look at the protocol and see if there are any suggestions we can make that may improve the results.

    According to the image sent the three anti phosphorilated GAB2 antibodies show different patterns, both #1 and #3 do not show any bands and #2 however gives several bands. Are the three antibodies targeting the same phosphorilated site in the protein?
