      Hi , I tried to use mouse sample this time and the antibody still cannot work. The attachment are two pictures taken from same membrane. The right three lanes in pic1 were bond with alpha3. The signal was very light. Using the same membrane, pic 2 shows staining with GABA receptor alpha1 antibody that we got it from Millipore. So, it is not because species, the antibody may have some problem. We purchased in May last year.

      1) What kind of samples were used (species and cell/tissue type)?
      We used this previous antibody on ferret brain tissue and mouse brain tissue for WB
      2) What dilution of the primary antibody was used?
      I used the dilution of 1:1000
      3) What blocking solution was used (BSA, milk, etc)?
      I used 5% milk for blocking.

      Thank you. I just want to you know I also use some other antibodies, for example anti-GABAA alpha 1, anti-GABAA gamma2 2, anti-GAD etc, the other antibodies work well on our ferret and mouse brain samples.

      Thank you for your reply. The anti-GABA A Receptor alpha 3 antibody, ab72446, is covered under our Abpromise and so is guaranteed to work in IHC-Fr on mouse tissue. If the antibody fails to perform as stated and if the scientific support team cannot help resolve this issue, then we would be happy to provide you with an alternative antibody or a refund. I have emailed the lab to request more information as the protocol used in testing ab72446 in IHC-Fr, and they have told me that the only information they have is what I have already sent you. . The lab did inform me that this antibody had been published in papers from Dr. Werner Sieghart lab and so searching his publications may yield finer protocol details. The protocol guides that I sent along yesterday should be able to provide you with more information on general IHC techniques Please let me know if there is anything else I can do to help. .

The only information I have been able to find about the IHC-Fr protocol use so far is that it was tested at 1/100 against on Rat amygdala (4% PFA + 15% picric acid fixed frozen sections.
