I received the attached flow cytometry protocol from the lab, which was used to validate ab176174. The laboratory did use a 2% PFA fixation, followed by permeabilization with 90% methanol, but this doesn't mean that a non-fixed population of cells permeabilized with a gentle detergent such as Tween 20 or saponin would not work, and I think it is worth attempting if you do not want to fix your cells.
