WB 1/1000. By Western blot, this antibody detects an 50 - 53 kDa protein, corresponding to the expected molecular weight of GABA A receptor beta 2 subunit.

    GABA, the major inhibitory neurotransmitter in the vertebrate brain, mediates neuronal inhibition by binding to the GABA/benzodiazepine receptor and opening an integral chloride channel.

Regarding the negative control, we do not have a KO mouse lysate. However, I would suggest checking the UniGene link as can be a good source to choose an appropriate positive or negative control.

I am sorry that we are not able to provide the sequence in this particular case. We are aware that this is not an ideal situation.Although we are providing the immunogen in many cases, this is not true in these circumstances.

However, both antibodies are not completely without any information regarding the immunogen: both are derived within the cytosolic loop of the rat protein:

ab8340- the immunogen can be found between AA327-451 of the rat protein,
ab4046- the immunogen can be found between AA328-450 of the rat protein



In addition, the article you kindly sent to us contains the following statement: "Lacking the antigen (e.g., if it is a large protein), there are several alternative strategies for establishing staining specificity." Thiskind of information can be found on the datasheetforab4046:

1. The antibody produces one single band

2. We provide next to the positive control brain the negative control in form of a ko-mice lysate, which is givenmore weight than the use of a blocking peptide.

Checking the datasheet, I found that ab4046has beenused six times in references from 2005 to 2012.ab8340 has one reference, but it is the only antibody against this target whichhas been cited.This shows that scientistsare successfully working and publishing with thesetwoantibodies.

I hope the information regarding the ab4046is therefore sufficient for the journal as it fits the required standards mentioned in the article. If this is not the case, please let me know.
