Target

  • Relevance
This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma aminobutyric acid from L glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin dependent diabetes. This gene may also play a role in the stiff man syndrome.
  • Cellular localization
Cytoplasm; cytosol. Associated to cytoplasmic vesicles. In neurons, cytosolic leaflet of Golgi membranes and presynaptic clusters.

  • Immunohistochemical analysis of formalin-fixed, paraffin-embedded Rat pancreatics sections labelling GAD65 & GAD67 with ab49832 at a concentration of 1/1000. The secondary used was a biotin peroxidase.

  • ab49832 staining human pancreas tissue sections by IHC-P.  Sections were formaldehyde fixed and subjected to heat mediated antigen retreival in 10mM citrate buffer prior to incubation with ab49832, diluted 1/2000, for 1 hour at 25°C.  An Alexa Fluor® 546 conjugated goat anti-rabbit antibody, diluted 1/200, was used as the secondary.

Answer

Thank you for contacting us. We recommend that you use this antibody for IHC-P at a dilution of 1/1000, incuabted overnight. You should first block the tissue with 10% normal serum plus 1% BSA for 1-2 hours to diminish background.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for contacting us.

We are happy to help customers find the most suitable products for their research purposes; however we are not specialized in this particular field and do not want to recommend a product that may be unsuitable. We encourage customers to consult the latest literature available through PubMed and other resources in order to find the most up-to-date information about their specific research interests.

All three of ourAnti-GAD65 + GAD67 antibodies are guaranteed to work in mouse tissues. Ab49832 and Ab11070 are both tested and guaranteed in IHC. I would be very happy to help you to find the best secondary antibody for use with either of these products. In order to determine this could you please let me know the detection system you plan to use?

I am sorry that I could not be more helpful regarding the best marker for your needs, but I hope that the available literature in this area can provide some clarification. Please do not hesitate to contact us again with other needs or with any questions about our products.

Answer

Thank you for contacting us.
I am sorry to hearthat you are encountering some problems with this antibody.

As discussed over the phone, I am sending the link to our antigen retrieval protocols:

https://www.abcam.com/index.html?pageconfig=resource&rid=11488

When using IHC-P, then heat mediated antigen retreival in 10mM citrate buffer was performed prior to incubation with ab49832.

Also, blocking with more BSA, e.g. 5% , can help reducing the non-specific background.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
