with mouse brain, GAD67 transgenic mice
detects GAD67 in cytoplasm, but no staining for GAD65
Antigen retrieval: none
Ab: 1/1000 o/n
blocking: 1% BSA, 0.01% TritonX
2nd: 549-conjugated, works well
suggestions: try AR (send protocols), use more BSA for blocking non-specific staining

I am sorry to hearthat you are encountering some problems with this antibody.
As discussed over the phone, I am sending the link to our antigen retrieval protocols:
When using IHC-P, then heat mediated antigen retreival in 10mM citrate buffer was performed prior to incubation with ab49832.
Also, blocking with more BSA, e.g. 5% , can help reducing the non-specific background.
