WB 1/1000. Detects a band of approximately 67 kDa (predicted molecular weight: 67 kDa).
IHC-P 1/500. Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

We don’t recommend this antibody for human in IHC. In our hands human tissues showed weak and non-specific staining.

  • Function

    Catalyzes the production of GABA.
  • Tissue specificity

    Isoform 3 is expressed in pancreatic islets, testis, adrenal cortex, and perhaps other endocrine tissues, but not in brain.
  • Involvement in disease

    Defects in GAD1 are the cause of cerebral palsy spastic quadriplegic type 1 (CPSQ1) [MIM:603513]. A non-progressive disorder of movement and/or posture resulting from defects in the developing central nervous system. Affected individuals manifest symmetrical, non-progressive spasticity and no adverse perinatal history or obvious underlying alternative diagnosis. Developmental delay, mental retardation and sometimes epilepsy can be part of the clinical picture.

  • GAD67 was immunoprecipitated from 0.35 mg of human brain lysate with ab213508 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab213508 at 1/500 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10,000 dilution.

    Lane 1:  Human brain lysate 10 µg (Input). 

    Lane 2: ab213508 IP in human brain lysate. 

    Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab213508 in human brain lysate.

    Blocking/Dilution buffer: 5% NFDM/TBST.
