    Galactose Assay Kit (Colorimetric/Fluorometric) (ab83382) provides a simple and convenient method for direct measurement of galactose levels in various biological samples such as serum, plasma and other body fluids, as well as food and growth media. In this assay, oxidation of galactose generates a product that reacts with the galactose probe to produce color (OD570nm) or fluorescence (Ex/Em 535/587 nm). Liquid samples can be tested directly without purification. The assay is fast, convenient and sensitive. It can also be used as a high throughput assay.

    This product is manufactured by BioVision, an Abcam company and was previously called K621 Galactose Colorimetric/Fluorometric Assay Kit. K621-100 is the same size as the 100 test size of ab83382.

    Galactose (C6H12O6 MW: 180.16 g/mol) is one of the major naturally occurring sugars, a monosaccharide that differs from glucose only by the configuration of the hydroxyl group at the carbon-4 position. Often present as an anomeric mixture of α-D-galactose and β-D-galactose, this monosaccharide exists abundantly in milk, dairy products and many other food types such as fruits and vegetables.

I'm wondering if this assay can tell a difference between galactose and galactose-1 phosphate. Can it also tell a difference between galactose and other epimers of glucose?

Answer

Ab83382 has some cross-reactivity with Galactose-1-phosphate but its extremely low. There is no cross-reactivity with glucose and insignificant cross-reactivity with sucrose has been noticed.

Thanks a lot for your reply.

Did you get some information in regards to minimal possible rate of the detection of galactosidase (more specific than only < 10 nmol/well that the datasheet indicates).

I think that I wasn't clear: the customer does not intend to measure enzyme activity. He measures galactosidase, but this is not free galactosidase. It is galastosidase that is bound to the protein. Will the kit ab83382 measure bound galactosidase?

I can confirm that this kit (ab83382: Galactose Assay Kit) will measure only galactose. The detection range which your customer has is not associated with this kit. All we mention on the product datasheet is that this kit is sensitive and quantifies the free galactose only.

The ab83382 assay is for galactose. There is not going to be any measurement of enzyme activity in this kit. This kit will measure the total amount of free galactose in your samples.

With one Galactose Assay Kit (ab83382) you are provided with enough reagents for a total of 100 tests. As discussed over the phone this does include all the control assays that need to be performed (so taking the 6x2 standard curve tests which need to be performed, this leaves you with a total of 88 tests left.


There is no 96 well plate provided with the kit. This allows you flexibility in choosing which detection system to use with the kit (fluorescence, black plates, clear bottom; luminescence, white plates; colorimeter, clear plate).The fluorometric assay is ˜10 fold more sensitive than the colorimetric assay.
