    Target

    • Relevance

      A member of the family of glycosphingolipids, monosialoanglioside acts as a receptor and antigen for cholera. GM1 ganglioside, one of the glycosphingolipids widely distributed in all tissues, occurs in highest concentrations in the central nervous system (CNS). It is primarily located in the outer surface of the mammalian cell's plasma membrane and in synaptic membranes of the CNS. GM1 ganglioside modulates a number of cell surface and receptor activities as well as neuronal differentiation and development, protein phosphorilation and synaptic function.

      ab23943 staining Ganglioside GM1 in African Green Monkey CV1 cells by Immunocytochemistry/ Immunofluorescence. The cells were fixed in 4% paraformaldehyde, permeabilised in 100% methanol and then blocked using 5% serum for 1 hour at room temperature. Samples were then incubated with primary antibody at 1/10 for 16 hours at 4°C. The secondary antibody used was a goat anti-rabbit IgG conjugated to Alexa Fluor® 488 (green) used at a 1/200 dilution. Counterstained with DAPI (1 ug/ml).

    Answer

    The reason that species is not mentioned on the datasheet is because the target is species independent. The target is Ganglioside GM1, a molecule composed of a glycosphingolipid (ceramide and oligosaccharide) with sialic acids linked on the sugar chain. The molecular formula of this lipid is C77H139N3O31.

    Therefore, as long as your samples have this molecule present, this antibody will be suitable.

    Thank you for your enquiry.

    I can confirm that ab23943 antibody is sold as whole antiserumt. Unpurified antibodies, such as those sold as whole antiserum, ascites or tissue culture supernatant will not have a concentration stated on the datasheet. Antibody concentration is usually determined by protein assay, and serum / ascites / tissue culture supernatant will contain a lot of other proteins, which means the antibody quantification would not be accurate.

    I can confirm that for whole antiserum, concentration of antibody is known to very between 1 - 10 mg/ml. Please note this conc is not the conc of specificIgG present in soluiton.

    Thank you for your enquiry. I can confirm these are different antibodies, which have been tested in different applications. Tested applications covered by our guarnatee are listed on teh online datasheets: ab23943  Anti-Ganglioside GM1 antibody is tested ICC-IF, not tested WB ab31119  Anti-Ganglioside GM1 antibody  is tested in WB but not ICC-IF   All tested application information is stated on our datasheets, and these are updated as soon as any new information is brought to our attention. In this case, as one antibody is tested in ICC-IF and the other in WB, I am sorry we would regrettably not be able to guarantee either of these antibodies for both applications.     Question

    I am looking for an AB to analyze natural killer cells in mouse liver. The commonly used AB for FACS for Mouse NK cells NK1.1-AK (Clon PK 136) reacts with the NK cells for frozen sections for the spleen and lymph node but not for the liver. In the literature one can find that NK cells in the mouse liver can be detected with anti-asialo GM1 from WAKO (Japan). Could you tell me if the above named AB (ab23941 or ab23943) can be used in IHC of mouse liver for the NK cells and if yes, if you have this AB in stock. Maybe there is an alternative you can recommend? The second question is in regards to an AB that can detect via IHC in the mouse liver a clot (in our case to intraportal injected nude mouse liver in human Langerhansche islands for diabetes therapy). In my opinion this would be for example an Ab which reacts with Fibrin but not with Fibrinogen. As far as I know this AB only exist against human fibrin. Thanks ever so much. --------- German version -------------------------- Ich suche Antikörper zur immunhistochemischen Darstellung von natürlichen Killerzellen in der Leber von Mäusen. Kann ich bitte Angaben erhalten, ob die oben genannten AK (ab23941 oder ab23943) zur immunhistologischen NK-Darstellung in der Maus geeignet sind und wenn ja, ob diese AK derzeit auch lieferbar sind. Haben Sie gegebenenfalls eine andere Empfehlung? Die zweite Frage betrifft einen Antikörper, der im histologischen Schnitt in der Mäuseleber einen Gerinnungs-Clot nachweisen kann (in userem Fall um intraportal in die nude-Maus-Leber injiziierte humane Langerhanssche Inseln zur Diabetestherapie). Aus meiner Sicht wäre das bspw. ein AK der mit Fibrin, aber nicht mit Fibrinogen reagiert. Soweit mir bekannt, gibt es einen solchen AK bisher aber nur gegen humanes Fibrin. Whilst the literature that you detail provides evidence that NK cells in the mouse liver can be detected using an anti-asialo GM1 antibody the two antibodies that you detail are yet to be tested using immunohistochemistry as an application. Therefore we could not guarantee them to work. With regards to your second question unfortunately we do not have an antibody that distinguishes between fibrin (clotted blood) and fibrinogen. The sole Fibrin monoclonal antibody in our catalogue also detects Fibrinogen. I am sorry that I cannot be of more assistance on this occasion.
