Our Abpromise guarantee covers the use of ab264157 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|IP||Use at 2-5 µg/mg of lysate.|
|IHC-P||1/2000 - 1/10000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
FunctionInhibits DNA replication by preventing the incorporation of MCM complex into prereplication complex (pre-RC). It is degraded during the mitotic phase of the cell cycle. Its destruction at the metaphase-anaphase transition permits replication in the succeeding cell cycle.
Sequence similaritiesBelongs to the geminin family.
Developmental stageAbsent during G1 phase, accumulates during S, G2, and M phases, and disappears at the time of the metaphase-anaphase transition.

Geminin was immunoprecipitated from 1mg of HeLa (Human epithelial cell line from cervix adenocarcinoma) whole cell lysate with ab264157 at 3 µg per reaction. Western blot was performed from the immunoprecipitate using a different Geminin antibody at 1 µg/ml.
Lane 1: ab264157 IP in HeLa whole cell lysate.
Lane 2: Control IgG.
Exposure time: 3 mins.
Cells prepared using NETN lysis buffer.
Immunohistochemical analysis of formalin-fixed, paraffin-embedded human breast carcinoma tissue labeling Geminin with ab264157 at 1/5,000 dilution. Detection: DAB.
Epitope retrieval with citrate buffer pH 6.0 is recommended for FFPE tissue sections.
