FunctionEnhances MAP3K1 oligomerization, which may relieve amino-terminal mediated MAP3K1 autoinhibition and lead to activation following autophosphorylation. May play a role in the regulation of vesicle targeting or fusion.
Tissue specificityHighly expressed in germinal center but not mantle zone B-cells. Also expressed in lung, brain and placenta and at lower levels in other tissues examined.
Sequence similaritiesBelongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. STE20 subfamily.
Contains 1 CNH domain.
Contains 1 protein kinase domain.
Cellular localizationCytoplasm. Basolateral cell membrane. Golgi apparatus membrane.

Paraffin-embedded human endometrial cancer tissue stained for Germinal Center Kinase/GCK using ab238843 at 1/500 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
HepG2 (Human liver hepatocellular carcinoma cell line) cells stained for Germinal Center Kinase/GCK (Green) using ab238843 at 1/166 dilution in ICC/IF, followed by Alexa Fluor®488-conjugated Goat Anti-Rabbit IgG (H+L) secondary antibody.
The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C. The nuclear counterstain is DAPI (Blue).
