    Please note that a mistake was made in reference 4 (Mesaeli et.al., J. Cell. Biol. 1999 Mar 8;144(5):857-68). The antibody used for immunohistochemistry on paraformaldehyde fixed tissues was the crude serum version of this antibody (Abcam ab290) and not Clontech's monoclonal as stated. This product is supplied in 25% glycerol. During freezeing and thawing some phase separation might occur - Please ensure that the solution is mixed thoroughly but GENTLY before use.

  • Relevance
    Function: Energy-transfer acceptor. Its role is to transduce the blue chemiluminescence of the protein aequorin into green fluorescent light by energy transfer. Fluoresces in vivo upon receiving energy from the Ca2+ -activated photoprotein aequorin.

    Subunit structure: Monomer.

    Post-translational modification: Contains a chromophore consisting of modified amino acid residues. The chromophore is formed by autocatalytic backbone condensation between Ser-65 and Gly-67, and oxidation of Tyr-66 to didehydrotyrosine. Maturation of the chromophore requires nothing other than molecular oxygen.

    Biotechnological use: Green fluorescent protein has been engineered to produce a vast number of variously colored mutants, fusion proteins, and biosensors. Fluorescent proteins and its mutated allelic forms, blue, cyan and yellow have become a useful and ubiquitous tool for making chimeric proteins, where they function as a fluorescent protein tag. Typically they tolerate N- and C-terminal fusion to a broad variety of proteins. They have been expressed in most known cell types and are used as a noninvasive fluorescent marker in living cells and organisms. They enable a wide range of applications where they have functioned as a cell lineage tracer, reporter of gene expression, or as a measure of protein-protein interactions. Can also be used as a molecular thermometer, allowing accurate temperature measurements in fluids. The measurement process relies on the detection of the blinking of GFP using fluorescence correlation spectroscopy.

    Fluorescent light microscopy analysis of a vertical retina cryosection (A) showing both CX3CL1/Cherry (detected in rhodamine channel, red) (B) and CX3CR1/GFP (detected in FITC channel, green) (C) reporters expression. I believe that ab6556 has not specifically been tested for reactivity with EYFP. However, this antibody is reactive against all variants of Aequorea victoria GFP such as S65T-GFP, RS-GFP, YFP and EGFP so I would expect that it would work.

This antibody is polyclonal, so it will recognise multiple epitopes on the GFP molecule. Being as the immunogen used was recombinant GFP, there is a very high chance that this antibody will recognise both the N-term and the C-term. It has been shown to cross react with all variants of Aequorea victoria GFP such as S65T-GFP, RS-GFP, YFP and EGFP. It has also been shown to work well in IP and WB.

The system you have chosen should be fine, although you may want to consider using a biotinylated secondary antibody followed by an ABC-peroxidase system. This is a more sensitive system because the biotinylation process is less harsh than conjugation. You should yield a stronger signal with an ABC method, too. Which ever you choose, remember to use in 10% normal goat serum made up in TBS with 1% BSA to block non-specific binding of the secondary. Also use 3% hydrogen peroxide to block endogenous peroxidases. If you choose to use an ABC method, consider incorporating an endogenous biotin block because kidney is a known to be biotin rich.
