    Please note that a mistake was made in reference 4 (Mesaeli et.al., J. Cell. Biol. 1999 Mar 8;144(5):857-68). The antibody used for immunohistochemistry on paraformaldehyde fixed tissues was the crude serum version of this antibody (Abcam ab290) and not Clontech's monoclonal as stated. This product is supplied in 25% glycerol. During freezeing and thawing some phase separation might occur - Please ensure that the solution is mixed thoroughly but GENTLY before use.

  • Relevance
    Function: Energy-transfer acceptor. Its role is to transduce the blue chemiluminescence of the protein aequorin into green fluorescent light by energy transfer. Fluoresces in vivo upon receiving energy from the Ca2+ -activated photoprotein aequorin.

    Subunit structure: Monomer.

    Tissue specificity: Photocytes.

    Post-translational modification: Contains a chromophore consisting of modified amino acid residues. The chromophore is formed by autocatalytic backbone condensation between Ser-65 and Gly-67, and oxidation of Tyr-66 to didehydrotyrosine. Maturation of the chromophore requires nothing other than molecular oxygen.

    Biotechnological use: Green fluorescent protein has been engineered to produce a vast number of variously colored mutants, fusion proteins, and biosensors. Fluorescent proteins and its mutated allelic forms, blue, cyan and yellow have become a useful and ubiquitous tool for making chimeric proteins, where they function as a fluorescent protein tag. Typically they tolerate N- and C-terminal fusion to a broad variety of proteins. They have been expressed in most known cell types and are used as a noninvasive fluorescent marker in living cells and organisms. They enable a wide range of applications where they have functioned as a cell lineage tracer, reporter of gene expression, or as a measure of protein-protein interactions. Can also be used as a molecular thermometer, allowing accurate temperature measurements in fluids. The measurement process relies on the detection of the blinking of GFP using fluorescence correlation spectroscopy.

    Fluorescent light microscopy analysis of a vertical retina cryosection (A) showing both CX3CL1/Cherry (detected in rhodamine channel, red) (B) and CX3CR1/GFP (detected in FITC channel, green) (C) reporters expression. I can reassure you that we undertook an in-depth study examining storage conditions and antibody performance. After over 13,000 individual ELISA experiments, we have determined that storing antibodies at various temperatures of up to 45°C for 1 week does not impact their activity.
Therefore, in this case I would not expect there to be significant impact on the quality of the antibody. I can suggest to try it in your experiments.

However, we do encourage customers to follow the long-term storage instructions on the datasheet as soon as they receive a product, so that it remains stable and so that we can continue to provide our guarantee.

Unfortunately, we cannot help you in this case: rabbits produce no subtypes of the IgG class (as they only have the C-gamma gene). If you would like to perform an isotype control, rabbit serum should do the trick.

I am sorry to confirm that these GFP antibodies have not been tested for detection of GFP from Zoanthus sp.

Our GFP antibodies, including ab6556, ab1218 and ab290are all raised against immunogens from the Aquorea victoria GFP protein, which has low alignment of just 19% with GFP from Zoanthus sp.Regrettably, this suggests that the antibodies would not detect GFP from Zoanthus sp.

There should be very little detection difference between the two but the laboratory has not tested this. YFP and GFP are different by only a few amino acids and the ab290 or ab6556 are very high titer polyclonal antibodies. So the producers of these antibodies think that they should recognize both FPs to practically the same extent.
