  • Function
    Ghrelin is the ligand for growth hormone secretagogue receptor type 1 (GHSR). Induces the release of growth hormone from the pituitary. Has an appetite-stimulating effect, induces adiposity and stimulates gastric acid secretion. Involved in growth regulation.
    Obestatin may be the ligand for GPR39. May have an appetite-reducing effect resulting in decreased food intake. May reduce gastric emptying activity and jejunal motility.
  • Tissue specificity
    Highest level in stomach. All forms are found in serum as well. Other tissues compensate for the loss of ghrelin synthesis in the stomach following gastrectomy.

    Belongs to the motilin family.
  • Post-translational
    modifications
    O-octanoylation or O-decanoylation is essential for ghrelin activity. The O-decanoylated forms Ghrelin-27-C10 and Ghrelin-28-C10 differ in the length of the carbon backbone of the carboxylic acid bound to Ser-26. A small fraction of ghrelin, ghrelin-28-C10:1, may be modified with a singly unsaturated carboxylic acid.
    Amidation of Leu-98 is essential for obestatin activity.

According to SwissProt, O-octanoylation or O-decanoylation is essential for Ghrelin activity. As this antibody detects both the octanoylated and non-octanoylated forms of Ghrelin, the antibody detects therefore both the active and form of the hormone. However, I am wondering if you are referring to the Ghrelin Receptors and not to the hormone? If this is the case, this antibody is unsuitable for you.
Regarding the Leptin antibody, because we carry over 70,000 products, it isn't feasible for us to keep small sample sizes of our products. Nonetheless, may I ask what kind of species you are working on?

Thank you for your patience. I am sorry to hear that you have been experiencing problems with ab19314 in western blot. Often it is possible to make suggestions that help resolve problems. We will happily offer technical support and in the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 120 days of purchase), and if it appears that the antibody is at fault, a free replacement/credit note/refund will be offered. I have looked through the protocols you used and have a few questions and suggestions that might help you resolve the problem. If the suggestions do not prove to be helpful, would you please be so kind to confirm the following items in order to help me better understand the cause of the problem. Can you confirm you have reduced and denatured the sample at 95oC for 10 minutes in buffer containing SDS and mercaptoethanol? The bands you are observing could be multimers and boiling at the proper temperature and duration will ensure that the protein is in the correct conformation to run at the correct molecular weight and be detected by the antibody. Have you done a protein assay to assess the concentration of protein you are loading onto the gel? We recommend loading 20 to 40ug of total protein per well. Loading more than this may overload the gel, which will result in many non-specific bands. Please try this amount if you have not already done so. Thank you for your enquiry. I contacted the originator of the antibody and there is no data regarding the detection of the unprocessed full-length precursor protein at this time. The antibody has been tested against recombinant human Ghrelin. It may detect the unprocessed precursor based on a BLAST search for sequence homology. Thank you for your enquiry. I have been in touch with the source of the antibodies that you are interested in and I can tell you that with regards to Anti-Ghrelin ab19314 this antibody was tested using recombinant human ghrelin. However, the highest expression has been found to be in stomach, which would make this a suitable control. Furthermore according to literature endocrine cells of the oxyntic mucosa have been used as a positive control by IHC when applying antibodies against this target. (ab19314 has only been tested by western blotting). ab15860 was tested against its immunising peptide by ELISA. However, again I have been informed that Ghrelin is expressed mostly in stomach and therefore mouse stomach tissue lysate can be used as positive control by western blotting. Unfortunately I have been unable to contact the source of the following antibodies; ab13982 and ab13984. However, given that they are against the same target I would recommend a similar positive control to the above.
