Target

  • Function

    Receptor for ghrelin, coupled to G-alpha-11 proteins. Stimulates growth hormone secretion. Binds also other growth hormone releasing peptides (GHRP) (e.g. Met-enkephalin and GHRP-6) as well as non-peptide, low molecular weight secretagogues (e.g. L-692,429, MK-0677, adenosine).
  • Tissue specificity

    Pituitary and hypothalamus.
  • Involvement in disease

    Defects in GHSR may be a cause of idiopathic short stature autosomal (ISSA) [MIM:604271]. Short stature is defined by a subnormal rate of growth.

    ab95259 Ghrelin Receptor/GHS-R antibody immunoreactivity in the mouse brain a.anterior hypothalamic nucleus b.high magnification immunoreactivity shows dendritic processes (arrowheads) and the periphery of the soma (arrows) of pyramidal cells in the somatosensory cortex c. hippocampus. The immunostaining with ab95250 is consistent with published data (Zigman et al, 2006).

    ab95250 staining GHS-R in SKNSH cells. The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated with the antibody ab95250 at 5µg/ml and ab7291 (Mouse monoclonal to alpha Tubulin - Loading Control) used at a 1/1000 dilution overnight at +4°C. The secondary antibodies were ab150081, Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed, (pseudo-colored green) and ab150120, Goat polyclonal Secondary Antibody to Mouse IgG - H&L (Alexa Fluor® 594) preadsorbed, (colored red), both used at a 1/1000 dilution for 1 hour at room temperature. DAPI was used to stain the cell nuclei (colored blue) at a concentration of 1.43 µM for 1hour at room temperature.

    Answer

    Antibody ab92520 is raised against a ghrelin receptor amino acid sequence that is found in both isoforms of the human receptor, so it should be capable of recognizing both. Antibody ab85104 is raised against a sequence derived from rat receptor that is 80% conserved in both human isoforms. It will probably recognize the human isoforms but the probability is lower than for ab92520.

    Was the sample deglycosylated before being run on the western blot? Also did you ever see any dimerization or multimerization of the protein when using this antibody?

    Answer

    Thank you for contacting Abcam about ab95250. I have heard back from the lab about the dimerization/glycosylation states that are recognized by the antibody, ab95250. The antibody has been tested on a number of different lysates which aren’t shown on the datasheet and in two (pituitary and small intestine) we saw no 37 kDa band but we did see week bands at ~55 and 70 kda. I think this is around the right region to see dimerisation, but as we don’t see a 37 kDa band my feeling is it was just a fail on those lysates, however ther is a chance that the antibody is atually recognizing the dimer states in these samples. Also, the samples were not deglycosylated before use. They are only reduced and denatured. Please let me know if there is anything else I can help you with.
