The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use a concentration of 16 µg/ml. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
ICC/IF Use at an assay dependent concentration.

  • Immunohistochemical analysis of formalin-fixed, paraffin embedded Human anterior pituitary tissue labeling GHRHR with ab150575 at 16ug/ml.
  • Immunofluorescence analysis of sheep anterior pituitary somatotropes, staining GHRHR with ab150575.

Cells were fixed with formaldehyde and blocked with 20% serum for 2 hours at 20°C; antigen retrieval was by heat mediation in a citrate buffer (pH 6). Samples were incubated with primary antibody (16 µg/ml in 2% normal goat serum in TBS) for 2 hours at 25°C. An AlexaFluor®488-conjugated goat anti-rabbit polyclonal IgG (2 µg/ml) was used as the secondary antibody.

Thank you for contacting us yesterday in regards to the anti-GHRHR antibody (ab150575).

Unfortunately, as you have discovered from the datasheet of this antibody, this antibody has not been used with sheep samples to our knowledge as yet. However, I have now checked the sequence homology shared between the immunogen used to raise the antibody and the sheep protein. This is 100% and we would therefore expect the antibody to cross react, however as we have not tested it we would not be able to guarantee this.

However, as discussed over the phone, if you would like to purchase ab150575 and use it with your sheep samples, we can offer you a testing discount. This offer means that if you purchase ab150575 as normal, test the antibody with your rabbit samples and let us know of the results through an Abreview (no matter whether positive or negative) you would then be eligible to claim the value of ab150575 (£227) off any product in our catalogue. More information on this offer can be found here:

Please note that the antibody to be tested must be purchased, tested, the Abreview submitted and the free product claimed within a 4 month period.

If you would be interested in participating in this scheme please do let me know as a discount code needs to be issued prior to the purchase of ab150575.

I hope this information has been of help. If you require any further information please do not hesitate to contact us again.
