    GLIS3 is a member of the GLI similar zinc finger protein family, and encodes a nuclear protein with five C2H2 type zinc finger domains. It functions as both an activator and repressor of transcription, and is specifically involved in the development of pancreatic beta cells, thyroid, eye, liver and kidney. Mutations in this gene have been associated with neonatal diabetes and congenital hypothyroidism (NDH). Alternatively spliced variants that encode different protein isoforms have been described but the full length nature of only two have been determined.

Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab51268. I would also appreciate if you can confirm some further details:
1. Have the cells you have been using been frequently passaged? If so, then these cells could gradually accumulate differences in their protein expression profiles so I would recommend using an original non-passaged cell line.
2. Did you use protease inhibitors? If not, use a protease inhibitor which will prevent the degradation of your protein.
3. Try boiling your samples for 10 minutes in SDS-PAGE to prevent the formation of multimers which could account for the band at 130 kDa.
Unfortunately, because I don't have an image for western blot using rat samples it is difficult to identify these extra bands present on yur blot. I have done a literature search and the only sequence for rat GLIS3 protein that I have found has not been reviewed and verified. I would expect to see a band at 100kDa as was seen by the customer who submitted the abreview. Unfortunately, I do not know whether this other customer also saw extra bands on the blot. On your blot, there appears to be a very faint band at 100kDa which I believe is the correct band.
From the literature, I have found that for mouse and human, other isoforms of this protein exists.
e.g. For mouse: isoform 2 is at 72 kDa, for human, isoform 2 is at 100 kDa whilst isoforms 1 are at ˜84 kDa.
It is likely that rat also has other isoforms which we do not yet know about and could be the reason for these extra bands.
I suggest trying out the suggestions above. If these do not improve your results, I will be happy to exceptionally apply the abpromise guarantee and offer you a free of charge replacement, credit note or refund.
