    Facilitative glucose transporter. This isoform likely mediates the bidirectional transfer of glucose across the plasma membrane of hepatocytes and is responsible for uptake of glucose by the beta cells; may comprise part of the glucose-sensing mechanism of the beta cell. May also participate with the Na(+)/glucose cotransporter in the transcellular transport of glucose in the small intestine and kidney.

  • ICC/IF image of ab54460 stained MCF7 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab54460, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Answer

The GLUT2 antibody ab54460 might be suitable for flow cytometry. We have not tested it in that application and our "Abpromise" guarantee of a replacement or refund in the event of failure does not apply to untested applications, but it has been tested for immunocytochemistry for microscopy, as seen at this link: https://www.abcam.com/glucose-transporter-glut2-antibody-ab54460.html#description_images_1.


Immunocytochemistry and flow cytometry use similar staining protocols, but different tools for collecting data: microscope versus flow cytometer. The antibody ab54460 detects an intracellular domain of GLUT2, so fixation and permeabilization will be required before staining, which means the antibody cannot stain live cells.


We do have one GLUT2 antibody that has been tested for flow cytometry, ab85715. Here is a link to the data: https://www.abcam.com/glucose-transporter-glut2-antibody-5d1-ab85715.html#description_images_3 .


Please note that it has only been tested with human cells. We don't know if it will detect cells of other species.
