Customer reviews and Q&As

Application
Flow Cytometry
Sample
Human Cell (Peripheral Blood Mononuclear Cells (PBMC))
Permeabilization
Yes - BD Cytofix/Cytoperm™ Fixation/Permeablization Kit
Gating Strategy
First, lymphocytes are gated based on FSC-A vs. SSC-A parameters. Then, single cells are gated by attending to FSC-A vs. FSC-H parameters. Next, dead cells are excluded with the viability dye
Specification
Peripheral Blood Mononuclear Cells (PBMC)
Preparation
Cell harvesting/tissue preparation method: Thawed PBMC have been cultured in RPMI (+10% FBS, +1% non-essential aa, +1% pyiruvate, +1% penicillin/streptomycin) for 3 days with IL-15 (1 ng/uL). Then, cells were collected in a conical tube, and washed with 5 mL PBS (centrifuge at 300g for 5 minutes).
Sample buffer: PBS
Fixation
BD Cytofix/Cytoperm™ Fixation/Permeablization Kit
