Target

  • Function

    Ionotropic glutamate receptor. L-glutamate acts as an excitatory neurotransmitter at many synapses in the central nervous system. Binding of the excitatory neurotransmitter L-glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist.
  • Involvement in disease

    Defects in GRIA3 are the cause of mental retardation X-linked type 94 (MRX94) [MIM:300699]. Mental retardation is characterized by significantly sub-average general intellectual functioning associated with impairments in adaptative behavior and manifested during the developmental period. MRX94 patients have moderate mental retardation. Other variable features are macrocephaly, seizures, myoclonic jerks, autistic behavior, asthenic body habitus, distal muscle weakness and hyporeflexia.

    Palmitoylated. Depalmitoylated upon glutamate stimulation. Cys-621 palmitoylation leads to Golgi retention and decreased cell surface expression. In contrast, Cys-847 palmitoylation does not affect cell surface expression but regulates stimulation-dependent endocytosis.

The datasheet is confusing because its difficult to determine if the antibody will specifically detect metaboropic or ionotropic glutamate receptor 3.

Thank you for your enquiry.

I have found that several years ago ab40845 was incorrectly listed as an antibody to metabotropic glutamate receptor 3. However it was then correct to list as ionotropic receptor. Unfortunately some indications from the old metabotropic datasheet survived, including those 2 citations and some alternative names.

So I can confirm ab40845 is specific for the ionotropic receptor. Also, the immunogen sequence is from amino acid 296 - 314 of the human protein.

I will contact our IT department to have the metabotropic related items removed from this datasheet to make things more clear.

Hi I would like to know if this antibody is selective for mGulR3 or if it cross reacts with mGluR2? Is it possible to have the peptide sequence so that I can check if this antibody is likely to cross react?

I'm afraid we are unable to provide the peptide sequence, however we have performed a sequence alignment of the immunogen with the mGluR2 protein and can confirm that ab40845 is not predicted to cross react with mGluR2 due to very low homology.
