    Catalyzes the attachment of the cognate amino acid to the corresponding tRNA in a two-step reaction: the amino acid is first activated by ATP to form a covalent intermediate with AMP and is then transferred to the acceptor end of the cognate tRNA.

      Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Glutamyl Prolyl tRNA synthetase/PARS antibody (ab31531)

      IHC image of ab31531 staining in human breast carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab31531, 1/1000 dilution, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

      ab31531 staining Glutamyl Prolyl tRNA synthetase/PARS in the Human Liver cell line Huh7 by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with PFA, permeabilized with Triton X-100 0.5% and blocked with 10% Serum for 1 hour at 25°C. Samples were incubated with primary antibody (1/300 in PBS + 10% Serum) for 1 hour at 25°C. An Alexa Fluor®488-conjugated Donkey anti-rabbit IgG polyclonal (1/500) was used as the secondary antibody.

    I am glad to see your newly improved product ab31531 (anti-human GluProRS antibody) in your website. Because one of my research project concerns this protein, I have some technique question here. 1. in your product datasheet, the predicted MW of GluProRS is 163 KD. I saw you guys used the old GluProRS datum (P07814) in which GluProRS lacks of 72 residues at N terminus, so it is not a real full-length GluProRS. You can find the updated datum of human GluProRS in NCBI protein databases, the locus is NP_004437. Newly published human GluProRS has 1512 amino acids and its predicted MW should be 172KD. 2.Our lab have tried to over-express and purify human GluProRS in several different system for a long time. But we could not get high level expression. I hope to know which expression system you did use to over-express human GluProRS and how to purify it. If it is possible, could you help us make an expression plasmid that has the full-length human GluProRS sequences (1512 aas)? The over-expressed protein should be easily purified from host cells. I am looking forward to hearing from you soon. And please send me an estimated cost for this service.
