Our Abpromise guarantee covers the use of ab83550 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use a concentration of 5 µg/ml.
ELISA Use at an assay dependent concentration.
WB Use a concentration of 1 µg/ml. Predicted molecular weight: 81 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.

  • Function

Transfers the glycosyl residue from UDP-Glc to the non-reducing end of alpha-1,4-glucan.
  • Pathway

    Glycan biosynthesis; glycogen biosynthesis.
  • Involvement in disease

Defects in GYS2 are the cause of glycogen storage disease type 0 (GSD0) [MIM:240600]; A metabolic disorder characterized by fasting hypoglycemia presenting in infancy or early childhood, high blood ketones and low alanine and lactate concentrations. Although feeding relieves symptoms, it often results in postprandial hyperglycemia and hyperlactatemia.
  • Sequence similarities

Belongs to the glycosyltransferase 3 family.
  • Post-translational

Primed phosphorylation at Ser-657 (site 5) by CSNK2A1 and CSNK2A2 is required for inhibitory phosphorylation at Ser-641 (site 3a), Ser-645 (site 3b), Ser-649 (site 3c) and Ser-653 (site 4) by GSK3A an GSK3B. Dephosphorylation at Ser-641 and Ser-645 by PP1 activates the enzyme.

  • Anti-Glycogen synthase 2 antibody (ab83550) at 1 µg/ml (in 5% skim milk / PBS buffer) + human fetal muscle lysate at 10 µg

HRP conjugated anti-Rabbit IgG at 1/50000 dilution

    Predicted band size: 81 kDa
    Observed band size: 81 kDa

    Gel concentration 12%
  • IHC image of ab83550 staining in Human skeletal muscle formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab83550, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Customer reviews and Q&As

Thanks for your call yesterday and for your patience while I have been in touch with the lab about ab83550. I've attached a lab image showing Western blot results in human liver samples.

We discussed our testing discount program, since we haven't tested ab83550 in frog samples at this time, and I've attached more information about this program below. Please let me know if you have any questions and I'll be happy to help!

Since ab83550 has not been tested in woodfrog, I can offer a discount off a future purchase if you buy ab83550 now, test it in woodfrog, and submit feedback to us in the form of an Abreview. The discount would be to the value of 1 free primary antibody.

If you are interested in this offer, please follow these steps:

1. Reply to this e-mail to let me know that you would like to proceed and test ab83550 in woodfrog. I will then send a discount code. This code must be issued before purchasing ab83550 so please wait for my reply before ordering.

2. Purchase ab83550 either by phone, fax, or online (www.abcam.com).

3. Test it in woodfrog.

4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews.

5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any primary antibody ordered and the discount code is valid for 4 months after issue.

We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if ab83550 turns out to be unsuitable for woodfrog, you will still receive the discount on your next purchase after your Abreview has been submitted.

Please let me know if you have any questions about this offer and I would be happy to help you further.
