WB Use at an assay dependent concentration. To detect mGM-CSF by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant mGM-CSF is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
ELISA Use at an assay dependent concentration. To detect mGM-CSF by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant mGM-CSF.
Neutralising Use at an assay dependent concentration. To yield one-half maximal inhibition [ND50] of the biological activity of mGM-CSF (0.6 ng/ml), a concentration of 0.012 - 0.02 µg/ml of this antibody is required.
IHC-P Use a concentration of 5 µg/ml.

  • Western blot analysis of recombinant GM-CSF using ab9741 at 0.1μg/ml. Bottom band - dye front.

  • Less than 0.10 ug/ml ab9741 will neutralize 50% of the bioactivity of 1 ng/ml m-GM-CSF.
  • IHC image of GM-CSF staining in Human normal tonsil formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab9741, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

The neutralizing and inhibition functions stated on the datasheet of the antibodies ab9741 andab64715 relate to the assays which have been performed to validate their activities.

With the Anti-GM-CSF antibody (ab9741), the antibody has been used in a neutralising assay as described in the protocol attached to this email.

Whilst with the Anti-TGF beta 1 antibody [2Ar2] (ab64715) has been used in an inhibition of proliferation assay. This was carried out asdescribed in Tsang, M. et al., (1995) Cytokine 7:389.The dose response curves are given in figures 1 and 2 of the attached document.The response is linear over a small range.

The immunogen of ab9741 was the whole GM-CSFprotein of mouse. Mouse GM-CSF only shows 71% homology with rat as a whole protein and therefore I cannot recommend to use this antibody for rat samples.

We do have two antibodies that have a immunogen (peptide) that is taken from a part of the GM-CSF human or ratsequence that shows a higher homology to the rat sequence. Both antibodies are predicted to react with rat and I would be happy to issue a discount code for either of them.

Maybe your customer would like to review the datasheets and choose one of them for testing in rat?

The GM-CSF protein is expected to run around 16 kDa, however there are 4 known glycosylation sites, each adding around 2 kDa to the protein, so the protein generally runs around 24 kDa on a Western blot.
