Thank you for contacting us for technical support and for sending us all this protocol information and an image, it is very useful to understand your problem. The antibody ab7113 has not been tested for DotBlot so we do not know if it will work in this application and we cannot guarantee it will. I think the problem you are having may be due to the blocking: I would recommend trying 5% milk or 5%BSA in TBST to block, only for 1hour, then add the primary and then the secondary antibodies (diluted in TBST only, no milk).
