      I have some questions for you. Recently, I tried to use your antibodies, ab49394(primary AB) and ab5929( 2nd antibody), doing IHC for measure macrophages of small intestine tissue of human for a few times. I got good stains for positive samples without background, but negative control(without primary antibody)also got some positive spots. I tried to block it, I used:
      1. 5% BSA in TBS with tween 20, 2 hours at RT;
      2. 5% normal goat serum in TBS, 2 hours at RT;
      3. I also blocked endogenous Avidin used vector Lab,SP-2002 and endogenous HRP used Dako, S2003.
      Could you give me some suggestions to solute this problem?

      Question

      I am working with B cells which express an human IgG or human IgM as well as a murine IgM. I would like to show the expression of the murine IgM in FACS. Therefore I need an antibody which ONLY recognizes the murine IgM but not the human IgM or IgG. In your catalogue I found an "Mouse IgM secondary antibody - µ chain speciic- F(ab)2 Fragment (ab5932). Do you know if this antibody crossreacts with human IgM/G? Unfortunately this antibody is coupled to PE. Since my cells are GFP positive I prefere Biotin or APC labeled antibodies. Do you have an anti mouse-µ antibody not reacting with human IgG/M which is coupled to Biotin or APC?

      Answer

      We do not have any APC labeled IgM antibodies. Other conjugates we have- Texas red, AP, HRP,FITC, Rhodamine. ab5929- Biotin labelled. Seems to be the best option as it is also tested in Flow cytometry. However, neither ab5929 or ab5932 is cross adsorbed or tested against human IgM, and we cannot guarantee their suitable for your assay. Two other biotin conjugated anti-mouse IgG antibodies but only tested in ELISA: ab11659 Mouse IgM secondary antibody [LO-MM-9] - Heavy Chain specific. Unable to guarantee non-reactivity with human Ig. ab49760 will not recognise other mouse Ig's as there was no reaction against IgA, IgG, and IgM. Human IgG or IgM cross-reactivity has not been tested.
