  • ICC/IF image of ab7291 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab7291, 1µg/ml) overnight at +4°C. The cells were then incubated with ab7075, rabbit anti-mouse HRP conjugated antibody, for 1h at a 1/250 dilution. The cells were then incubated with ab34580, guinea pig anti-HRP antibody, for 1h at a 1/250 dilution. Lastly, the secondary antibody (orange) was ab150188 Alexa Fluor® 594 goat anti-guinea pig IgG (H+L) used at 1µg/ml for 1h, as a quaternary antibody. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Application
Immunohistochemistry (PFA perfusion fixed frozen sections)
I used Free floating LDT mouse brain slices fixated in 4% PFA. The slices were 40µm thick. Permeabilization was with 150µLTritonx and blocking with 2% goat serum in 50mL PBS for 30 min. I used two primary antibodies (poly. rabbit anti-bNOS and poly. Guinea pig anti-GABA). 1/400 ratio. The were incubated at room temp. for 24 hrs. I used two secondary antibodies (Alexa 488 goat anti-rabbit and Alexa 594 goat anti-guinea pig). The bNOS stain was fabulous but the GABA stain was bad. There was bad background and non-specific staining everywhere.

Abcam have spoken to this customer and have suggested that the poor results are probably attributed to the primary antibody rather than to the secondary antibody. We have therefore given the customer protocol tips to improve his results with the primary antibody.
