Answer

Thank you for providing that extra information.

We have previously used the Anti-CD4 antibody [CT-4] (ab25345) in flow cytometry at a concentration of 0.1 ug per 106 cells. Meanwhile the secondary antibody ab97239 can be used in a range of 1/50 to 1/200 to start off with.

These are only suggested as starting points and may require further optimisation with your particular samples and protocol.

I hope this information has been of help. If you have any further questions, please do not hesitate to ask.

Both the goat polyclonal anti-mouse IgG (ab97239 and ab98692) should be compatible with the mouse anti-CACNA1H antibody (ab84815) for the experiment you hope to perform. The antibody ab98692 is provided pre-adsorbed. This can be very useful when using multi-color imaging and IHC experiments when several primary antibodies and their corresponding secondary antibodies are used simultaneously. It also reduces the risk of cross-reactivity between the secondary antibody and endogenous immunoglobulins present on cell and tissue samples. It can therefore help to reduce non-specific staining.

I hope this information has been of help. If you have any further questions, please do not hesitate to ask.

Answer

Thank you for your enquiry and your interest in our products.
I have conducted a search for you and found several antibodies against Sarcomeric Alpha Actinin. One of them is ab9465 which has been tested in ICC/IF and it recognizes cardiac muscle in different species (Mouse, Rat, Sheep, Rabbit, Goat, Chicken, Hamster, Cow, Cat, Dog, Pig, Xenopus laevis, Fish, Lizard, Snake, Xenopus tropicalis ). We have excellent feedback (3 Abreviews) from our customers who have used it and also specific publications (11). As you can see on the databases, it is a mouse monoclonal antibody and the isotype is IgG1.
For selecting the correct compatible secondary antibodies, I would advise you to use our advanced search engine: https://www.abcam.com/index.html?pageconfig=productmap&cl=918
You can type the target and the host species, isotype, format, specificity, conjugate etc. Have you considered the fluorochromes you wish to use?
Here are a few suggestions:
ab97239: Goat anti-Mouse IgG1 heavy chain (FITC) secondary antibody,
ab98692: Goat polyclonal Secondary Antibody to Mouse IgG1 - heavy chain (FITC), pre-adsorbed
ab72553: Goat polyclonal Secondary Antibody to Mouse IgG1 (SureLight® Allophycocyanin)
Regarding ICC protocol, I would advise you to take a look at the Protocol section at our website. We have a great numbers of general protocols which may be useful for you:

Thank you for your enquiry and interest.

You could certainly use our advanced search engine specifically designed for secondary antibodies to find the most suitable products for your studies:

https://www.abcam.com/index.html?pageconfig=productmap&cl=918

Since the isotype of this primary antibody is mouse IgG1, so you could select the isotype, target species, host species, format, specificity, conjugate using the drop-down menu.

We have different conjugates such as FITC, DyLight488, Chromeo 488, and here are a few suggestions:

FITC:

ab98692

ab97239

ab11588

You can find some further information about the the generation of our fluorochromes at this site:https://www.abcam.com/index.html?pageconfig=resource&rid=12948

DyLight488:

Affinity purified: ab96875, ab97015, ab96871

Pre-adsorbed: ab96879

Chromeo 488:

ab60313

Answer

1. yes,you need a secondary antibody such as ab97239 (Goat anti-Mouse IgG1 heavy chain (FITC) secondary antibody). The reason isthat the primary antibodies you want to use (ab25345, ab25346 andab25347) are not conjugated.

2. a.Yes, ab98693 (Goat anti-Mouse IgG1 heavy chain (HRP) secondary antibody)is compatible with the Mouse IgG1 primary antibodies you want to use.

2. b. As shown on the datasheet https://www.abcam.com/ab98693, ab98693 has not been used in IHC-Fr yet. However, it has been tested in IHC-P, so it is very likely that it willwork in IHC-Fr.
