    IHC image of Histone H1.0 staining in human colon formalin fixed paraffin embedded tissue section*. The section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with ab11080, 7.5µg/ml overnight at +4°C. An HRP-conjugated secondary (ab97240, 1/2000 dilution) was used for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

    The inset negative control image is secondary-only at 1/500 dilution.

    Super specific 2nd antibody against mouse IgG1 Heavy Chain only

    Western blot
    This antibody is super specific! It does not react with mouse IgG2a or human IgG, or any IgG light chain I tested so far.

    I use it because I work with mouse tumor lysate in western blot. All tumor lysate contains blood, so IgG is always in the lysates. When my protein of interest is at the same molecular weight of IgG light chain, I use this antibody.

    The purpose of western blot I show here is to verify if this antibody reacts with light chain of mouse IgG1 specifically, so there is no primary antibody used.

    The antibody is quite clean. The image I show is over-exposed with some background in order to confirm there is absolutely no IgG light chain signal.

    I have some specifictechnical questions regarding your secondary antibodies:

    1) What level of cross-reactivity is there between thefollowing four antibodies(ie how specific for their IgG subclass target):

    Thank you for your interest in our secondary antibodies. I will answer your questions as best I can now but cross-reactivity of the mouse anti-human IgG isotype antibodies will require data from tests which I do not have at hand. I will forward these to you when I receive them.

    For the second question, each antibody of each pair is the same with regards to isotype specificity but differ with regards to species specificity. Please note that you have reversed the two anti-IgG1 antibodies in your list: ab98693 is pre-adsorbed for species reactivity, and ab97240 is not.

    anti IgM ab97230 vs ab98679 - Same with respect to isotype specificity. (No reactivity with other isotypes was detected). Cross-adsorbed to remove reactivity with human and rat IgG. (Less than 1% reactivity).

    anti IgG1 ab98693 vs ab97240 - ab98693 is pre-adsorbed for species reactivity, and ab97240 is not. As above, regarding isotype specificity and species reactivity.

    anti IgG2a ab97245 vs ab98698 - As above, regarding isotype specificity and species reactivity.

    anti IgG2b ab97250 vs ab98703 - As above, regarding isotype specificity and species reactivity.

    Thank you for your enquiry.

    I can confirm that ab43ERCC1 antibody [3H11] is a mouse monoclonal,IgG2a isotype.

    The ab97240 secondary antibody detects mouse IgG1 isotype, so it would not be suitable for use with ab43. You will need a secondary antibody that either specifically detects mouse IgG2a, or one that detects the more general IgG isotype.

    I would be pleased to help find some more suitable secondaries foryou. In order to do so, I would appreciate if you could confirm which application you will be using (for example, WB, IHC-P). I can then find an antibody suitable for your experiments.

I am looking for anti-mouse IgG2a and IgG1 secondary antibodies (either HRP- or AP- conjugated) to use for Isotyping of specific mouse serum IgGs against my desired antigen. I searched through your products but I could not find any product. I should say that the secondary antibodies I need must not cross-react with other mouse IgG subtypes. There are three different IgG1 and three different IgG2a antibodies which have been tested against other isotypes.
