Thank you for your inquiry. Indeed, antibodies as most (unmodified) proteins have a carboxy group (-COOH) at their C-terminus, as you indicated. They also have an amine group (-NH2) at their N-terminus. Furthermore, antibodies contain amino acids which have carboxy and amine groups in their side-chains, such as Arginine, Lysine, Glutamate or Aspartate. Therefore, the coupling reaction with FITC that usually takes place via free amine groups forming a stable thiourea bond (as for example with our EasyLink kits) is absolutely feasible for the anti-Enrofloxacin/Ciprofloxacin antibody ab21035. However, as the Goat polyclonal Secondary antibody ab97230 is already conjugated to HRP, I would not recommend to label this antibody with FITC. As alternative, I would suggest using an unconjugated antibody such as ab98671: Click here (or use the following: https://www.abcam.com/index.html?datasheet=98671). Thank you for your reply. Both antibodies suggested in my last email (ab98671 and ab98743) that were suitable for ELISA should also detect IgM in denatured or reduced forms when being used as primary antibody in WB. We also offer ab98679 which is HRP labelled and could be used directly without need for a secondary antibody that then is HRP labelled. In this case it is not necessary to use an F(ab’)2 antibody.
