  • ICC/IF image of ab8227 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab8227, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab150089 Alexa Fluor® 488 goat anti-rabbit IgG (Fc) used at 2µg/ml for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

La promoción para testar el Rabbit Monoclonal consiste en que al pedir cualquier primario, se os regala un Rabbit Monoclonal. Lamentablemente contra GFP el único rabbit monoclonal que hay solo se ha testado en WB. Al no tener pruebas de su uso en otras aplicaciones, no puedo asegurarte que vaya a funcionar.

Si no te sugiero que pidáis el anti GFP testado en IF y el Rabbit Monoclonal contra otra proteína que necesitéis detectar.

Respecto a los secundarios, la diferencia entre ambos es la fracción del primario que reconocen. El ab150077 reconoce tanto la cadena pesada como la cadena ligera del anticuerpo, mientras que ab150089 solo detecta la fracción pesada del primario. El primero permite una señal más intensa al unirse al mayor número de primarios posibles, mientras que el segundo genera menos background al no poder unirse a ningún sitio antigénico. Cualquiera de ellos es válido para inmunofluorescencia.
