  • IHC image of beta Actin staining in normal human colon, formalin-fixed and paraffin-embedded tissue*. The section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with ab8227, 3µg/ml overnight at +4°C. An HRP-conjugated secondary (Ab97200, 1/200 dilution) was used for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

    The inset negative control image is taken from an identical assay without primary antibody.

The secondary antibodies you mentioned might work for dot blots with the rabbit polyclonal Histone primaries, as they are guaranteed to work in Western blot which is quite similar to dot blot. However, we have not tested them specifically for this application and thus cannot guarantee that it would work.

Therefore, I would like to recommend one of the following secondaries which have been tested in dot blot:

They are raised in goat, donkey and sheep and apart from this are equally well suited for your experimental setup. If you need a decision guiding you could for example choose the one with the most references: this would be ab6721 with 52 publications mentioning the use of this antibody.

We have been informed by the source of the secondary antibodies ab97195 and ab97200 that : By immunoelectrophoresis and ELISA, ab97195 (anti-Rabbit IgM - HRP) reacts specifically with rabbit IgM. This antibody may cross react with IgM from other species.   By immunoelectrophoresis and ELISA,  ab97200 (anti-Rabbit IgG - HRP) reacts specifically with rabbit IgG-Fc. This antibody may cross react with IgG from other species.
