Electron Microscopy Use at an assay dependent dilution.

  • Double immuno-labeling of UGT8 and ceramide in stably transfected PC-3 (Human prostate adenocarcinoma cell line) cells.
    Two different secondary antibodies conjugated to different sizes of gold nano-particles (ab105277; 12 nm, labels ceramide, red arrows) and (ab27234; 10 nm, labels UGT8, yellow open arrows). More ceramide labeling and less UGT8 labeling is observed in sham and down regulation samples and the opposite is observed in the up-regulation samples. UGT8 labeling was observed in both the Golgi and the endoplasmic reticulum; both are sites for ceramide metabolism.

    Incubated with secondary antibodies at a 1/50 dilution for 45 mins at room temperature.

The attachment of gold particles to ab27234 Goat Anti-Rabbit IgG H&L (10nm Gold) is by passive adsorption, not via a covalent bond.

We do have kits that will allow one to covalently attach gold particles to antibodies. These are ab154873 and ab154876.

The optical density of gold conjugates of this particle size at 520nm wavelength is 3.0. This allows the conjugates to be used in a highly diluted form. The appropriate dilution will vary between applications so the only way to establish the appropriate dilution for a specific application is to create a dilution range and see which dilution gives the best results.
