    By immunoelectrophoresis and ELISA this antibody reacts specifically with rabbit IgG and with light chains common to other rabbit immunoglobulins. No antibody was detected against non-immunoglobulin serum proteins. Less than 0.1% cross reactivity to bovine, chicken, horse, human, mouse, pig, and rat IgG was detected. This antibody may cross react with IgG from other species.

  • ICC/IF image of ab6046 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab6046, 1µg/ml) overnight at +4°C. The secondary antibody (yellow) was ab175695 Alexa Fluor® 568 Goat anti-Rabbit IgG (H&L) used at 2µg/ml for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

     

    Alexa Fluor® 568 is pseudocolored in yellow.

Immunocytochemistry/ Immunofluorescence
ICC/IF image of ab175695 Goat anti-rabbit IgG conjugated with Alexa Fluor 568 stained Jurkat cells.

The cells were ethanol+acetic acid fixed (10 min), permabilised in 0.4% Tween-20 for 30 min and incubated with the primary antibody CD45 (ab40763), 1 in 200 dilution, overnight at 4 degree C. abcam blocking buffer was used to block non-specific protein-protein interactions. Secondary antibody was Alexa Fluor® 568 as shown in red. DAPI was used to stain the cell nuclei (blue).

Additional Notes The image was taken by compact fluorescent cell imaging system, ZOE from Bio-Rad. ZOE, an android operated system, is easy to use and allows user to merge double stains in one click with good image quality.
