    The section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH 6) for 30mins. The section was incubated with ab8227, 3 µg/ml overnight at +4°C. An HRP-conjugated secondary (ab97051, 1/2000 dilution) was used for 1hr at room temperature. The section was counterstained with hematoxylin and mounted with DPX.

Thank you for your quick response. If you would be willing to send us a replacement vial, that would be great.
I was looking at alternative anti-rabbit IgG (HRP) antibodies and noticed that ab6721 had the same description as ab97051.
What is the difference between these two products? I'm tempted to use ab6721 instead since it has a number of references
associated with it. Is there any reason we can't use ab6721 with ab65166? If ab6721 can be used with ab65166, I'd like
to give that a try; if it is not compatible with ab65166, then another lot of ab97051 would be appreciated.

Inquiry: Hello, I recently purchased ab97051 from you all and have tried to do some dot blots using this secondary antibody. For these experiments, we have been directly spotting brain (mouse) whole cell lysate on PVDF membrane (one spot 10 ug, one spot 2 ug). In our first experiment, we tried blocking with 5% milk for one hour at room temperature. We then incubated with the primary antibody (ab65166) at three different dilutions (1:500, 1:1000, and 1:2000) overnight at 4 oC. We subsequently incubated with 1:10,000 of ab97051 for one hour at room temperature. After incubation with each antibody, we did three five-minute washes with TBS-Tween, and, at the end, a final wash with TBS. The blot was visualized using an Amersham ECL+ kit. All our blots had incredibly high background with white dots where we spotted the protein against a black background. We tried washing the blots overnight in TBS but still saw similar results. We tried the experiment again blocking instead with 10% milk overnight at 4 oC and using a 1:25,000 of the secondary. We also did one blot that was not incubated with the primary antibody. All the blots (including the one not exposed to primary antibody) gave white spots on a black background even when exposed for very short periods of time (<10 seconds). Our interpretation of these results is that there is a high degree of non-specific binding to both the PVDF membrane and to the cell lysate. However, we also wanted to make sure that we were employing appropriate blocking conditions. Have you all found that 10% milk overnight at 4 oC is sufficient to block non-specific binding of the antibody?

Thank you for getting back to me and for specifying the sample types.
As you have kindly explained to me in your previous e-mail, the primary and the secondary antibodies worked with the positive control but the signal was very weak using lysates from sheep cardiac tissues.
This antibody (ab3583) should recognize sheep Leptin, the cross-reactivity with this species has been tested and confirmed.
After reading through the detailed protocol you kindly forwarded to Abcam, I would like to make the following comments/suggestions:
1) Lysis buffer:
Leptin is secreted so potentially it can be degraded very quickly. It is very important to prepare the samples from fresh tissues and use lysis buffer which contains proteinase inhibitors. The whole process should be carried out on ice. I understand from the initial e-mail that Laemmli buffer was used for lysing the samples. I would rather suggest using RIPA buffer to prevent any unwanted protein degradation in the sheep cardiac tissues. The recipe of RIPA can be found at this website:

2) Loading amount:
Try to load at least 25-30 ug total tissue lysate per lane onto the gel.
3) Dilution:
You may need to optimize the dilutions of the primary antibody to find out the most suitable working dilution range for sheep cardiac tissue lysate. Brain tissue has high levels of leptin but cardiac tissue may have significantly lower. I have tried to find some information for you regarding the expressed levels of leptin in sheep tissue, but unfortunately not too much data are available on the public sites compared to human, rat our mouse (i.e. Swiss-Prot, Unigen).

The expression levels of leptin may vary from cell type to cell type or from tissue to tissue. It would be worth testing 1/500 or 1/1000 (for 1 hr at room temperature or overnight at 4oC).
