The proteolytic cleavage at S1 site generates an extracellular subunit and a seven-transmembrane subunit. Furin is involved in the cleavage of the S2 site generating a soluble fragment. Processing at the GPS domain occurred independent of and probably prior to the cleavage at the S2 site.

Immunohistochemical analysis of formalin-fixed, paraffin embedded Human placenta tissue labeling GPCR GPR126 with ab218046 at 1/200 dilution, followed by conjugation to the secondary antibody and DAB staining.
Immunohistochemical analysis of formalin-fixed, paraffin embedded mouse liver tissue labeling GPCR GPR126 with ab218046 at 1/600 dilution, followed by conjugation to the secondary antibody and DAB staining.
After antigen retrieval, endogenous peroxidase was blocked by 3% hydrogen peroxide for 30 minutes. Blocking buffer (normal goat serum) at 37°C for 20 minutes. Antibody incubation with ab218046 was done overnight at 4°C.
