    • Immunocytochemistry/Immunofluorescence analysis of HeLa (human cervix adenocarcinoma) cells labelling GPCR LGR6 with purified ab126747 at 1/500. Cells were fixed with 100% methanol. ab150077, Alexa Fluor® 488-conjugated goat anti-rabbit IgG (1/1000) was used as the secondary antibody. Nuclei were counterstained with DAPI (blue).

      Secondary Only Control: PBS was used instead of the primary antibody as the negative control.

    • Overlay histogram showing LoVo cells stained with ab126747 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab126747, 1/100 dilution) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was rabbit IgG (monoclonal) (1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

    I aligned the zebrafish sequence with the human sequence and there was only a 59% level of homology, between the 2. I also aligned the actual immunogen sequence used for ab126747, and the level of homology for that particular sequence was about 50%.

    Based on this information, I do not think that any of the lgr6 antibodies that we have will react with Zebrafish.
