    Target

    • Function

Lysine demethylase that demethylates both histones and non-histone proteins. Enzymatically inactive by itself, and becomes active following phosphorylation by PKA: forms a complex with ARID5B and mediates demethylation of methylated ARID5B. Demethylation of ARID5B leads to target the PHF2-ARID5B complex to target promoters, where PHF2 mediates demethylation of dimethylated 'Lys-9' of histone H3 (H3K9me2), followed by transcription activation of target genes. The PHF2-ARID5B complex acts as a coactivator of HNF4A in liver. PHF2 is recruited to trimethylated 'Lys-4' of histone H3 (H3K4me3) at rDNA promoters and promotes expression of rDNA.
    • Tissue specificity

Widely expressed, including in liver (at protein level).
    • Sequence similarities

Belongs to the JHDM1 histone demethylase family. JHDM1D subfamily.
      Contains 1 JmjC domain.
      Contains 1 PHD-type zinc finger.
    • Domain

The PHD-type zinc finger mediates the binding to H3K4me2 and H3K4me3.
    • Post-translational
      modifications

Phosphorylated by PKA on specific serine residues, leading to the formation of an active lysine demethylase complex.
    • Cellular localization

      Nucleus > nucleolus.

      Question

      Hi,
We are interested in purchasing a PHF2 antibody. Your antibody ab65771
seems to be promising, but we have some questions about the peptide used
to produce the antibody. The issues are, a 269 aa peptide is very very
big for a synthetic peptide; is the information online correct? And,
both human and mouse PHF2 have 1096 amino acids, while on the
information on your webpage indicates the synthetic peptide used to
generate the ab "corresponding to C terminal amino acids 830-1098 of
Human GRC5 / PHF2". If possible please let us know if any of these
information is incorrect, and provide us the correct information.
      Thank you,

      Answer

Thank you for contacting us and for your patience while I have been in touch with the lab regarding your enquiry.
You are correct that there was a mistake on the datasheet. The immunogen runs from amino acid 830 to the C-terminus at amino acid 1096. It may be more accurate to call this a fragment than a peptide.
I have updated the datasheet with the correct immunogen information, but please let me know if you have any further questions or if there is anything else that we can do for you.
      Have a nice day!
