    Has chemotactic activity for neutrophils. May play a role in inflammation and exerts its effects on endothelial cells in an autocrine fashion. In vitro, the processed forms GRO-alpha(4-73), GRO-alpha(5-73) and GRO-alpha(6-73) show a 30-fold higher chemotactic activity.

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human placental tissue section (4µm) labelling GRO alpha with ab89318. Tissue was treated with 15µg/ml Protenase K for antigen retrieval and fixed with 4% PFA. A HRP-conjugated anti-mouse IgG was used as the secondary antibody (30 minutes at room temperature) followed by DAB color development.

Our neutralizing antibodies are tested for activity in vitro cell culture and unfortunately we have no information regarding the behavior of ab89318 for intraperitoneal or intratumoral injection in vivo. Our products are not designed for in vivo use and would not be guaranteed for those purposes. It is possible that this antibody may be suitable for your studies, and I agree that in theory you should be able to use less antibody by injecting directly into the tumor.

Thank you for your email. Iam sorry to hearthat this antibody is proving difficult to use in your experiment.

1. There are inherentdifficulties with antibody detection of recombinant tagged proteins that I can recommend considering. This may be a biological artifact rather than a problem with the antibody. Could you confirm that the protein transfected is full length and includes the immunogen sequence?

Since the recombinant GRO alpha is tagged, it is possible that the tag is preventing access to the antibody. Could you confirm at which region of the protein the tag has been placed?

2. Could you confirm the species from which the recombinant GRO alpha gene was taken?

3. I can recommend that due to the points described above, it is important to use an endogenous positive control sample. For example, thisprotein is highly expressed inliver tumor cells, lung tumor cells and chondrosarcomacells.

4. When testing our antibodies, our lab uses 5% BSA as a blocking reagent, so I recommend switching to this instead of milk. Some antibodies give stronger, more specific signals on blots blocked with BSA instead of milk, so doing this may improve the results you are seeing. An example of the above is the western blot obtained with the Abcam GAPDH antibody ab9385 : https://www.abcam.com/ab9385 .
