WB Use a concentration of 1 µg/ml. Predicted molecular weight: 63 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 5 µg/ml.

  • Function
    Cleaves the alpha-chain at multiple sites and the beta-chain between 'Lys-53' and 'Lys-54' but not the gamma-chain of fibrinogen and therefore does not initiate the formation of the fibrin clot and does not cause the fibrinolysis directly. It does not cleave (activate) prothrombin and plasminogen but converts the inactive single chain urinary plasminogen activator (pro-urokinase) to the active two chain form. Activates coagulation factor VII.

  • Anti-HABP2 antibody (ab81490) at 1 µg/ml (in 5% skim milk / PBS buffer) + MCF7 cell lysate at 10 µg

    HRP conjugated anti-Rabbit IgG at 1/50000 dilution

    Predicted band size: 63 kDa
    Observed band size: 63 kDa
    Additional bands at: 27 kDa, 28 kDa. We are unsure as to the identity of these extra bands.

  • IHC image of HABP2 staining in Human normal liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab81490, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
